Spike mutation pipeline reveals the emergence of a more transmissible form of SARS-CoV-2 Declaration of Interests: The authors declare no competing interests. Summary We have developed an analysis pipeline to facilitate real-time mutation tracking in SARS-CoV-2, focusing initially on the Spike (S) protein because it mediates infection of human cells and is the target of most vaccine strategies and antibody-based therapeutics. To date we have identified fourteen mutations in Spike that are accumulating. Mutations are considered in a broader phylogenetic context, geographically, and over time, to provide an early warning system to reveal mutations that may confer selective advantages in transmission or resistance to interventions. Each one is evaluated for evidence of positive selection, and the implications of the mutation are explored through structural modeling. The mutation Spike D614G is of urgent concern; it began spreading in Europe in early February, and when introduced to new regions it rapidly becomes the dominant form. Also, we present evidence of recombination between locally circulating strains, indicative of multiple strain infections. These finding have important implications for SARS-CoV-2 transmission, pathogenesis and immune interventions. Scientists say a now-dominant strain of the coronavirus appears to be more contagious than original ... The new strain appeared in February in Europe, migrated quickly to the East Coast of the United States and has been the dominant strain across the world since mid-March, the scientists wrote. In addition to spreading faster, it may make people vulnerable to a second infection after a first bout with the disease, the report warned. The 33-page report was posted Thursday on BioRxiv, a website that researchers use to share their work before it is peer reviewed, an effort to speed up collaborations with scientists working on COVID-19 vaccines or treatments. That research has been largely based on the genetic sequence of earlier strains and might not be effective against the new one. The mutation identified in the new report affects the now infamous spikes on the exterior of the coronavirus, which allow it to enter human respiratory cells. The report's authors said they felt an "urgent need for an early warning" so that vaccines and drugs under development around the world will be effective against the mutated strain. ...

The Effect of Chloroquine, Hydroxychloroquine and Azithromycin on the Corrected QT Interval in Patients with SARS-CoV-2 Infection ... Results - Two hundred one patients were treated for COVID-19 with chloroquine/hydroxychloroquine. Ten patients (5.0%) received chloroquine, 191 (95.0%) received hydroxychloroquine and 119 (59.2%) also received azithromycin. The primary outcome of TdP was not observed in the entire population. Baseline QTc intervals did not differ between patients treated with chloroquine/hydroxychloroquine (monotherapy group) vs. those treated with combination group (chloroquine/hydroxychloroquine and azithromycin) (440.6 ± 24.9 ms vs. 439.9 ± 24.7 ms, p =0.834). The maximum QTc during treatment was significantly longer in the combination group vs the monotherapy group (470.4 ± 45.0 ms vs. 453.3 ± 37.0 ms, p = 0.004). Seven patients (3.5%) required discontinuation of these medications due to QTc prolongation. No arrhythmogenic deaths were reported. Conclusions - In the largest reported cohort of COVID-19 patients to date treated with chloroquine/hydroxychloroquine {plus minus} azithromycin, no instances of TdP or arrhythmogenic death were reported. Although use of these medications resulted in QT prolongation, clinicians seldomly needed to discontinue therapy. Further study of the need for QT interval monitoring is needed before final recommendations can be made.

Flynn, the interviewing agents told McCabe, "had a very good recollection of events, which he related chronologically and lucidly," did not appear to be "nervous or sweating," and did not look "side to side" -- all of which would have been "behavioral signs of deception." McCabe wrote that Flynn seemed "completely normal" -- even when, on three occasions, Flynn looked at the window and told agents, "What a beautiful black sky." McCabe maintained that Flynn made that memorable comment three times -- first, at "noon," then an hour later, and then one more time shortly after that. However, McCabe's timeline in the book appeared to contradict the sentencing memorandum filed late last year by Flynn's attorneys, citing government documents. These teams of government agents are charged with asking questions, including about household members’ health, and extracting blood from all the people living in the homes. The reason given for the home visits is — you may have guessed it — coronavirus.J. Scott Trubey writes at the Atlanta Journal-Constitution that the government agencies are seeking the blood to test for “antibodies to the novel coronavirus to pinpoint who might have had COVID-19 and estimate how widely the virus has traveled.”People who live at the 420 randomly selected homes are free, Trubey writes, to refuse the questioning and blood drawing by the government inquisition and phlebotomy teams that show up at their front doors. But, in reality, people often find it hard to muster the courage to say “no” to government agents who accost them in person “asking” them to comply. People are intimidated. They think that even if they say “no” the requested action will still be taken anyway plus they will suffer additional consequences for resisting. That aids police efforts both to get people to say incriminating things and to obtain “permission” to search people and property, even from people who know evidence of a crime is likely to be found.A CNN report by Dakin Andone shows that the government teams knocking on doors are employing some of the look of cops in their effort to obtain maximum voluntary compliance. Uniforms, government badges, official letters are all part of the process, just like when cops show up for a home search holding a warrant. Andone writes: “Health workers conducting the survey have CDC vests and badges, the news release said, and are carrying a letter from the CDC and the Georgia Department of Public Health.” ...

  8. let's go back in time a bit to... 1968. in '68 we lived through what became known as the Hong Kong Flu Pandemic, or H3N2. it originated in China (Wuhan coincidentally enough) and lasted until '69/'70. during that time, 1mm people worldwide died, with 100,00 of them in the US. back then we didn't close schools, shutter people to their homes, ruin the economy and just generally induce a fear in the populace. hell, we had what can be considered the greatest get together of all time - Woodstock. back then men were men, women were women and sheep were sheep and we knew the difference. today, the snowflake gene has taken over, not just here in the states but across the globe. you have people clamoring with a, 'wear your face mask to protect me from you', 'we must track all people, all of the time' and, 'we can't open society, we will go extinct if we do' mindsets, with little evidence to support that mantra. we are still learning what exactly this thing is and probably will for some time. there are many metaphors to describe what we are doing but two that come to mind are, 'shooting oneself in the foot' and 'cutting off your nose to spite your face'. time is going to tell just how disastrous the preventative cure (ruining the economy (where potentially millions are going to die because of)) will have been.
